Sharpie Oil Based Markers can be used on plastic. The traditional alcohol based Sharpie Markers are not ideal for plastic. They will smear and ultimately fade over time.
Use permanent markers on the plexiglass if you're drawing something you want to keep or writing something you'd like to be permanent. Place a white sheet of paper under the plexiglass to give yourself improved visibility so you don't make a mistake.
The cool thing about chalk markers is that you can use them on other surfaces too! They write on windows, glass, windshields, and more. You can even use them as a dry erase marker on dry erase boards, acrylic, and glass!

We use dry erase markers and grease pencils on plexiglass at work (Air Traffic Control Tower) and it comes up pretty good. Just watch out for scratches on the plexi; the marker likes to stick in there.
